Özet

Pepper (Capsicum annuum L.) is one of the most widely grown and economically important vegetable groups in the world. The primary factors limiting production inputs in open fields and greenhouses where pepper is cultivated are virus infections. Major infections in pepper growing areas are caused on by the PMMoV, one of the Tobamovirus group’s viruses that can spread mechanically and by seeds, and the ToBRFV, which has recently caused serious issues with tomato production. It is quite difficult to diagnose these two viruses symptomatologically, requiring RT-PCR with virus-specific primers for complete diagnosis. The usage of resistant cultivars is the best defense against viral infections. In this study, the results of MAS analyze performed with both classical PCR and qPCR using various markers identifying the L4 gene in pepper plants were compared. The markers used to identify the L4 gene were compared with the results of the MAS analysis. Additionally, a traditional PCR marker was identified, which enhances RealTime PCR and supports the observed results. As long as there are laboratory facilities available, it has been revealed that L4 classical markers should be confirmed using qPCR kits and that the results from using only one marker are unreliable.
