Comparison of Cytotoxic Effects of Essential Oil Obtained from Origanum majorana (L.) Plant on Epidermoid Carcinoma Cells (A431)

Özet

Background/aim: In recent years, attention has been drawn to the increasing incidence of skin

cancer. As a result, interest in higher plants with antioxidant capacity is increasing, as they can

delay the progression of many chronic diseases. The cytotoxic effect of Origanum majorana

essential oil on epidermoid carcinoma (A431) cells was measured by two different viability

tests and the results were compared.

Materials and methods: A431 (Epidermoid Carcinoma) cell line was obtained from the

Molecular Biology and Genetics Laboratory of Akdeniz University, Faculty of Science, in

frozen form. Cells were routinely cultured in Dulbeccos Modified Eagles Medium (DMEM),

supplemented with 10% fetal calf serum, and 1% antibiotic-antimycotic solution in atmosphere

containing 5% CO2 at 37 C. For subculturing, cells were harvested after a trypsin/EDTA

treatment at 37 °C and plated into 96-well plates. The 24-hour cytotoxic effect of Origanum

majorana essential oil on cells were demonstrated by the Cell Titer-Blue® cell viability and

MTT tests.

Results: In our study, the IC50 values of the cytotoxic effect of Origanum majorana essential

oil on epidermoid carcinoma (A431) cells with MTT and Cell Titer-Blue® cell viability test

were found to be 75 μg/ml and 74 μg/ml respectively.

Conclusion: This study showed that the essential oil of Origanum majorana exhibited a

cytotoxic effect due to the increased concentration. The results of both cytotoxic tests measuring

viability are very close to each other, confirming the results.
